Abstract

Latar Belakang: Staphylococcus aureus merupakan bakteri patogen yang dapat
menyebabkan berbagai jenis infeksi, sehingga memerlukan pengobatan dengan
antibiotik yang efektif, seperti Chloramphenicol. Uji sensitivitas antibiotik, seperti
metode difusi cakram Kirby-Bauer, penting untuk menilai efektivitas antibiotik
melalui pengamatan zona hambat di sekitar cakram. Mengingat pentingnya media
penyerapan, kertas saring seperti Whatman yang memiliki kemampuan serap
serupa dapat menjadi alternatif pengganti kertas cakram berdasarkan efisiensi
biaya. Tujuan: Penelitian ini bertujuan untuk mengetahui efektivitas penggunaan
kertas whatman sebagai alternatif pengganti kertas cakram pada uji sensitivitas
bakteri Staphylococcus aureus terhadap antibiotik Chloramphenicol. Metode:
Penelitian ini bersifat deskriptif dengan pendekatan cross sectional. Sampel dalam
penelitian ini berjumlah 12 sampel diambil dengan teknik purposive sampling.
Hasil: Hasil uji sensitivitas menunjukkan bahwa dari 12 sampel Staphylococcus
aureus yang diuji dengan antibiotik kloramfenikol menggunakan kertas Whatman,
sebanyak 91,67% tergolong sensitif dan 8,33% intermediet. Seluruh sampel
menunjukkan sensitivitas 100% pada kertas Whatman Grade 1 dan 42, sementara
25% sampel pada Grade 41 menunjukkan kategori intermediet. Berdasarkan
konsentrasi antibiotik, kloramfenikol pada 500 ppm, 750 ppm, dan 1000 ppm
menghasilkan daya hambat 100%, sedangkan pada 250 ppm, 66,7% sampel sensitif
dan 33,3% lainnya intermediet Kesimpulan: Didapatkan distribusi kertas whatman
sebagai alternatif pengganti kertas cakram pada uji sensitivitas bakteri
Staphylococcus aureus terhadap antibiotik Chloramphenicol sebesar 91,67% dalam
kategori sensitif dan 8,33% dalam kategori intermediet. Saran: Kepada masyrakat,
peneliti dan instansi pendidikan disarankan menggunakan kertas whatman grade 1
dan grade 42 dengan konsentrasi antibiotik lebih dari 250 ppm sebagai alternatif
pengganti kertas cakram.

Background: Staphylococcus aureus is a pathogenic bacterium that can cause
various types of infections, requiring treatment with effective antibiotiks such as
Chloramphenicol. Antibiotik sensitivity testing, such as the Kirby-Bauer disk
diffusion method, is essential to assess antibiotik effectiveness through the
observation of inhibition zones around the disks. Considering the importance of
absorbent media, filter paper such as Whatman, which has similar absorbent
properties, can be a cost-efficient alternative to commercial antibiotik disks.
Objective: This study aimed to determine the effectiveness of using Whatman filter
paper as an alternative to standard antibiotik disks in sensitivity testing of
Staphylococcus aureus against Chloramphenicol. Method: This research is
descriptive with a cross-sectional approach. A total of 12 samples were collected
using purposive sampling techniques. Results: The sensitivity test results showed
that out of 12 Staphylococcus aureus samples tested with Chloramphenicol using
Whatman paper, 91.67% were categorized as sensitive and 8.33% as intermediate.
All samples showed 100% sensitivity when using Whatman Grade 1 and 42 paper,
while 25% of the samples using Grade 41 paper were categorized as intermediate.
Based on antibiotik concentration, Chloramphenicol at 500 ppm, 750 ppm, and
1000 ppm showed 100% inhibitory activity, whereas at 250 ppm, 66.7% of samples
were sensitive and 33.3% were intermediate. Conclusion: The use of Whatman
filter paper as an alternative to commercial antibiotik disks in the sensitivity testing
of Staphylococcus aureus to Chloramphenicol showed 91.67% sensitivity and
8.33% intermediate response. Suggestions: It was recommended that the public,
researchers, and educational institutions consider using Whatman Grade 1 and
Grade 42 filter paper with antibiotik concentrations above 250 ppm as an
alternative to commercial antibiotik disks..
