Abstract
Latar Belakang : Pewarnaan sediaan apus darah tepi (SADT) metode Giemsa
memerlukan larutan fiksasi untuk menjaga morfologi sel darah. Metanol absolut
yang umum digunakan memiliki risiko toksik tinggi. Oleh karena itu, alkohol 96%
dipertimbangkan sebagai pilihan yang lebih aman karena bersifat kurang toksik,
lebih mudah diperoleh, dan umum digunakan dalam prosedur fiksasi lain di
laboratorium. Tujuan Penelitian : Diketahuinya apakah alkohol 96% dapat
menggantikan metanol absolut sebagai alternatif fiksasi pewarnaan sediaan apus
darah tepi (SADT) metode Giemsa. Metode Penelitian : Penelitian ini bersifat
eksperimental. Teknik sampling yang digunakan adalah random sampling. Sampel
penelitian diambil sebanyak 20 sampel. Penelitian ini dilaksanakan di Laboratorium
Jurusan Teknologi Laboratorium Medis Poltekkes Kemenkes Palembang. Analisis
data yang digunakan adalah uji Chi-Square. Hasil : Hasil menunjukkan bahwa
penggunaan alkohol 96% memberikan hasil morfologi eritrosit yang sebanding
dengan metanol absolut, dengan eritrosit normositik dan normokrom pada seluruh
sampel. Namun, bentuk eritrosit (krenasi) menunjukkan hasil sedikit lebih baik
pada metanol absolut dibanding alkohol 96% dengan uji Chi-Square diperoleh nilai
p value 0,451 > α (0,05). Karena nilai p > 0,05 maka keputusan uji Ho gagal ditolak
sehingga disimpulkan tidak ada pengaruh penggunaan larutan fiksatif metanol
absolut dan alkohol 96% pada pewarnaan sediaan apus darah tepi (SADT) metode
Giemsa terhadap morfologi eritrosit (ukuran, warna, bentuk). Kesimpulan :
Alkohol 96% dapat digunakan sebagai alternatif pengganti metanol absolut untuk
fiksasi pewarnaan SADT metode Giemsa dalam pemeriksaan morfologi eritrosit.
Background : Staining of peripheral blood smears (PBS) using the Giemsa method
requires a fixation solution to preserve the morphology of blood cells. Commonly
used absolute methanol has a high risk of toxicity. Therefore, 96% alcohol is
considered a safer option because it is less toxic, easier to obtain, and commonly
used in other fixation procedures in the laboratory. Research Objective : It is
known whether 96% alcohol can replace absolute methanol as a staining fixation
solution of the Giemsa method of Peripheral Blood Smear (PBS) Preparation.
Research Method : This research is experimental. The sampling technique used is
random sampling. The research sample was taken as many as 20 samples. This
research was carried out at the Laboratory of the Department of Medical
Laboratory Technology, Polytechnic of the Ministry of Health, Palembang. The
data analysis used was the Chi-Square test. Results : Results showed that the use
of 96% alcohol gave erythrocyte morphological results comparable to absolute
methanol, with normocytic erythrocytes and normochromes in the entire sample.
However, the erythrocyte form (crenasation) showed slightly better results in
absolute methanol than in 96% alcohol with the Chi-Square test obtaining a p value
of 0.451 > α (0.05). Because the p value > 0.05, the Ho test decision was rejected
so that it was concluded that there was no effect of the use of absolute methanol
fixative solution and 96% alcohol on the staining of Giemsa's Edge Blood Smear
Preparation (SADT) on erythrocyte morphology (size, color, shape). Conclusion :
96% alcohol can be used as an alternative to absolute methanol for fixation in the
staining of peripheral blood smears (PBS) using the Giemsa method in the
examination of erythrocyte morphology.
