Abstract
Latar Belakang: Escherichia coli merupakan bakteri patogen yang dapat
menyebabkan infeksi serius, terutama di negara tropis seperti Indonesia. Resistensi
antibiotik menjadi tantangan dalam penanganan infeksi ini, sehingga diperlukan
alternatif antibakteri dari bahan alami, salah satunya buah nanas varietas Queen
yang diketahui mengandung senyawa aktif seperti bromelain, flavonoid, dan
saponin. Tujuan Penelitian: Mengetahui efektivitas antibakteri jus buah nanas
(Ananas comosus (L) Merr Var. Queen) terhadap Escherichia coli dengan variasi
konsentrasi 25%, 50%, 75%, dan 100%. Metode Penelitian: Penelitian ini
menggunakan metode quasi-eksperimental dengan rancangan Posttest-Only
Nonequivalent Control Group Design. Jus buah nanas diuji terhadap Escherichia
coli menggunakan metode difusi cakram pada media Mueller Hinton Agar, dengan
enam pengulangan pada tiap perlakuan. Hasil Penelitian: Hasil menunjukkan
bahwa seluruh variasi konsentrasi jus nanas (25%, 50%, 75%, 100%) tidak
menghasilkan zona hambat terhadap Escherichia coli (diameter 0 mm). Sementara
itu, kontrol positif menggunakan Amoksisilin membentuk zona hambat dengan
diameter rata-rata 17,4 mm. Kesimpulan: Jus buah nanas varietas Queen tidak
menunjukkan aktivitas antibakteri terhadap Escherichia coli pada konsentrasi dan
metode yang digunakan. Hal ini dapat disebabkan oleh pengaruh pH jus yang terlalu
asam (pH 3,0) serta kompleksitas dinding sel bakteri gram negatif.
Background: Escherichia coli is a pathogenic bacterium that can cause serious
infections, especially in tropical countries like Indonesia. The growing issue of
antibiotic resistance necessitates the exploration of natural antibacterial
alternatives, such as pineapple (Ananas comosus (L) Merr Var. Queen), which is
known to contain active compounds like bromelain, flavonoids, and saponins.
Objective: To determine the antibacterial effectiveness of Queen variety pineapple
juice (Ananas comosus (L) Merr Var. Queen) against Escherichia coli at
concentrations of 25%, 50%, 75%, and 100%. Methods: This quasi-experimental
study used a posttest-only nonequivalent control group design. Pineapple juice was
tested against E. coli using the disk diffusion method on Mueller Hinton Agar, with
six repetitions for each treatment. Results: All tested concentrations of pineapple
juice (25%, 50%, 75%, 100%) produced no inhibition zone (0 mm) against
Escherichia coli. In contrast, the positive control using Amoxicillin formed a clear
inhibition zone with an average diameter of 17,4 mm. Conclusion: Pineapple juice
of the Queen variety showed no antibacterial activity against Escherichia coli under
the tested concentrations and methods. This may be due to the acidic pH (3.0) of
the juice and the complex structure of gram-negative bacterial cell walls.
