Abstract

Latar belakang : Media kultur merupakan gold standard dalam pemeriksaan
bakteri, namun media komersial harganya mahal dan hanya bisa ditemukan di
tempat tertentu. Alternatif dari bahan alam terus dikembangkan, termasuk dari
limbah organik. Rambut jagung (Zea mays L) mengandung nutrisi seperti
karbohidrat dan protein yang dibutuhkan bakteri, namun belum banyak
dimanfaatkan dalam mikrobiologi. Tujuan Penelitian : Untuk mengetahui
pemanfaatan rambut jagung (Zea mays L) sebagai media alternatif untuk
pertumbuhan bakteri Escherichia coli. Metode Penelitian : Penelitian ini bersifat
kuasi eksperimental dengan rancangan penelitian Post Test Only Non-Equivalent
Control Group Design. Penelitian ini dilakukan di Laboratorium Bakteriologi
Poltekkes Kemenkes Palembang Jurusan Teknologi Laboratorium Medis pada
bulan Febuari-Mei 2025. Hasil Penelitian : Koloni pada media alternatif memiliki
karakteristik bentuk bulat, keadaan permukaan halus, elevasi cembung dan ukuran
yang relatif sama dengan koloni pada endo agar. Pada pigmentasi koloni terdapat
perbedaaan dimana konsentrasi 5% dan 10% koloni bewarna merah muda,
sedangkan pada konsentrasi 15% dab 20% koloni bewarna putih. Kesimpulan:
Rambut jagung dapat dimanfaatkan sebagai media alternatif untuk pertumbuhan
bakteri Escherichia coli. Saran: Rambut jagung dapat digunakan sebagai media
alternatif dalam pemeriksaan kultur dan resistensi mikrobakteriologi.

Background : Culture media are essential for supporting bacterial growth;
however, commercially available media are often expensive and not readily
accessible. As a result, natural and low-cost alternatives, including those derived
from organic waste, are increasingly being explored. Corn silk (Zea mays L)
contains essential nutrients such as carbohydrates and proteins that are required
for bacterial growth, yet its application in the field of microbiology remains limited.
Objective : This study aims to evaluate the potential use of corn silk (Zea mays L)
as an alternative culture medium for the growth of Escherichia coli. Method : This

research employed a quasi-experimental design using the Post-Test Only Non-
Equivalent Control Group Design. The study was conducted at the Bacteriology

Laboratory of the Department of Medical Laboratory Technology, Poltekkes
Kemenkes Palembang, from February to May 2025. Results : Colonies grown on
the alternative medium were round, with smooth margins, convex elevation, and
comparable size to those observed on Endo agar. In terms of pigmentation, colonies
appeared pink at 5% and 10% concentrations, while at 15% and 20%
concentrations, colonies were white. Conclusion: Corn silk has the potential to be
utilized as an alternative medium for supporting the growth of Escherichia coli.
Suggestion: Corn silk can be used as an alternative medium in culture examination
and antimicrobial resistance testing in microbiology..
