Abstract
Latar Belakang : Infeksi yang disebabkan oleh Staphylococcus aureus semakin
sulit ditangani karena resistensi terhadap antibiotik. Penggunaan bahan alami
sebagai alternatif antibakteri menjadi pilihan yang potensial. Bawang putih (Allium
sativum) mengandung allicin yang bersifat antibakteri, sedangkan daun pepaya
(Carica papaya L) mengandung senyawa seperti alkaloid dan flavonoid yang juga
efektif menghambat pertumbuhan bakteri. Tujuan Penelitian : Penelitian ini
bertujuan untuk mengetahui efektivitas kombinasi ekstrak bawang putih (Allium
sativum) dan daun pepaya (Carica papaya L) sebagai antibakteri terhadap
pertumbuhan Staphylococcus aureus. Metode Penelitian : Penelitian dilakukan
secara in vitro menggunakan metode difusi cakram (Kirby-Bauer) dengan lima
variasi konsentrasi kombinasi: 20%, 40%, 60%, 80%, dan 100%. Sebagai kontrol
digunakan Amoxicillin (positif) dan aquadest steril (negatif). Hasil Penelitian :
Hasil penelitian menunjukkan bahwa kombinasi ekstrak bawang putih dan daun
pepaya pada semua konsentrasi tidak menunjukkan aktivitas antibakteri, yang
ditunjukkan oleh tidak adanya zona hambat. Sebaliknya, kontrol positif
menghasilkan zona hambat rata-rata sebesar 38 mm. Kesimpulan : Kombinasi
ekstrak bawang putih dan daun pepaya tidak efektif dalam menghambat
pertumbuhan Staphylococcus aureus pada metode dan konsentrasi yang digunakan.
Beberapa faktor yang mempengaruhi efektivitas antibakteri antara lain interaksi
antar senyawa, stabilitas senyawa dan strain bakteri yang digunakan.
Background: Infections caused by Staphylococcus aureus are increasingly difficult
to treat due to rising antibiotic resistance. The use of natural ingredients as
alternative antibacterial agents is a promising approach. Garlic (Allium sativum)
contains allicin, known for its antibacterial properties, while papaya leaves (Carica
papaya L) contain compounds such as alkaloids and flavonoids that are also
effective against bacterial growth. Objective: This study aimed to evaluate the
antibacterial effectiveness of a combination of garlic and papaya leaf extracts
against Staphylococcus aureus. Methods: An in vitro study was conducted using
the disk diffusion method (Kirby-Bauer) with five concentration variations (20%,
40%, 60%, 80%, and 100%). Amoxicillin and sterile distilled water were used as
positive and negative controls, respectively. Results: The combination of garlic and
papaya leaf extracts at all tested concentrations showed no antibacterial activity,
as indicated by the absence of inhibition zones. In contrast, the positive control
(Amoxicillin) produced an average inhibition zone of 38 mm. Conclusion: The
combination of garlic and papaya leaf extracts was not effective in inhibiting
Staphylococcus aureus under the conditions and concentrations used. Several
factors affect antibacterial effectiveness, including compound interactions,
compound stability, and the type of bacterial strain used.
