Abstract

Latar belakang: Potato Dextrose Agar (PDA) merupakan salah satu media yang
digunakan untuk pertumbuhan jamur Candida albicans. Media PDA dibuat di
pabrik dalam bentuk sediaan siap pakai, harganya mahal, higroskopis, dan hanya
diperoleh pada tempat tertentu. Nasi aking mengandung karbohidrat yang dapat
digunakan sebagai pengganti sumber karbohidrat pada media PDA. Tujuan
penelitian: untuk menganalisis pemanfaatan media nasi aking sebagai alternatif
pengganti media Potato Dextrose Agar (PDA) terhadap pertumbuhan Candida
albicans. Metode penelitian: penelitian ini merupakan penelitian quasi
eksperimental dengan desain penelitian posttet-only nonequivalent control group
design. Hasil penelitian: didapatkan hasil rerata pertumbuhan jamur Candida
albicans dengan menghitung jumlah dan diameter koloni pada hari ke5 pada
media PDA sebagai kontrol yaitu sebesar 54,5 CFU/mL dan 5 mm, lalu pada
media nasi aking konsentrasi 5%, 10%, 15%, 20% dan 25% yaitu sebesar 82,25
CFU/mL dan 1 mm, 96,5 CFU/mL dan 1 mm, 104,75 CFU/mL dan 1 mm, 78,75
CFU/mL dan 1 mm, 115 CFU/mL dan 1 mm. Hasil uji statstic menunjukkan tidak
ada pengaruh antara pertumbuhan jumlah koloni pada media PDA dan media nasi
aking dan adanya pengaruh antara pertumbuhan diameter koloni pada media PDA
dan media nasi aking. Kesimpulan: dapat disimpulkan bahwa media alternatif
nasi aking konsentrasi 5%, 10%, 15%, 20% dan 25% dapat digunakan untuk
mengganti media PDA dalam pertumbuhan Candida albicans yang memiliki
peluang untuk menumbuhkan jamur Candida albicans, namun tidak seefektif
media PDA dalam mendukung perkembangan ukuran koloni. Saran: disarankan
untuk menggunakan konsentrasi yang lebih tinggi ataupun menggunakan jamur
dengan spesies yang berbeda.

Background: Potato Dextrose Agar (PDA) is one of the media commonly used for
the growth of Candida albicans. PDA is produced in ready to use form by
factories, is expensive, hygroscopic, and only available in certain places. Nasi
aking (dried leftover rice) contains carbohydrates that can be used as an
alternative carbohydrates source to replace PDA. Research Objective: to analyze
the use of nasi aking as an alternative medium to replace Potato Dextrose Agar
(PDA) for the growth of Candida albicans. Research Method: this study used a
quasi-experimental design with a posttest-only nonequivalent control group
design. Research Results: the growth of Candida albicans was observed by
counting the number and measuring the diameter of colonies after five days of
incubation. On PDA media (control), colony growth was 54,5 CFU/mL with a 5
mm diameter. On nasi aking media at concentrations of 5%, 10%, 15%, 20%, and
25%, colony growth was 82,25 CFU/mL and 1 mm, 96,5 CFU/mL and 1 mm,
104,75 CFU/mL and 1 mm, 78,75 CFU/mL and 1 mm, 115 CFU/mL and 1 mm,
respectively. Statistical test showed no significant difference in the number of
colonies between PDA and nasi aking media. However, there was a significant
difference in colony diameter. Conclusion: nasi aking medium at concentrations
of 5%, 10%, 15%, 20%, and 25% can be used as a substitute for PDA in growing
Candida albicans, with potential for fungal growth. However, it is less effective
than PDA in supporting colony size development. Suggestion: it is recommended
to used higher concentrations or to experiment with fungi of different species.
