Abstract
ABSTRAK
Latar Belakang: Kelelahan merupakan kondisi menurunnya kemampuan tubuh dalam melakukan aktivitas fisik yang dapat memengaruhi produktivitas. Penggunaan tonikum sintetik secara terus-menerus berpotensi menimbulkan efek samping sehingga diperlukan alternatif dari bahan alam yang lebih aman. Daun jambu bol (Syzygium malaccense) diketahui mengandung berbagai senyawa metabolit sekunder yang berpotensi memiliki aktivitas tonikum. Penelitian ini bertujuan untuk mengetahui aktivitas tonikum ekstrak etanol daun jambu bol terhadap mencit putih jantan (Mus musculus).
Metode: Penelitian ini menggunakan metode eksperimental dengan rancangan pre-test dan post-test. Hewan uji dibagi menjadi lima kelompok yang terdiri atas kontrol negatif (Na-CMC 0,5%), kontrol positif (kafein), serta tiga kelompok perlakuan ekstrak etanol daun jambu bol dosis 50 mg/kgBB, 100 mg/kgBB, dan 200 mg/kgBB. Aktivitas tonikum diuji menggunakan metode natatory exhaustion dengan mengukur waktu bertahan renang mencit sebelum dan sesudah perlakuan selama 14 hari. Data dianalisis menggunakan uji Paired Sample t-Test, One Way ANOVA, dan uji lanjut Tukey.
Hasil: Hasil skrining fitokimia menunjukkan bahwa ekstrak etanol daun jambu bol mengandung alkaloid, flavonoid, tanin, saponin, dan steroid, sedangkan terpenoid tidak terdeteksi. Hasil analisis statistik menunjukkan adanya perbedaan yang signifikan antar kelompok perlakuan (p<0,05). Ekstrak etanol daun jambu bol mampu meningkatkan waktu bertahan renang mencit, dengan aktivitas tonikum terbaik ditunjukkan pada dosis 200 mg/kgBB yang menghasilkan peningkatan waktu renang sebesar 246,09%.
Kesimpulan: Ekstrak etanol daun jambu bol (Syzygium malaccense) memiliki aktivitas tonikum terhadap mencit putih jantan (Mus musculus). Dosis 200 mg/kgBB merupakan dosis yang paling efektif dalam meningkatkan daya tahan fisik mencit berdasarkan parameter waktu bertahan renang.

Background: Fatigue is a condition characterized by a decreased ability of
the body to perform physical activities, which can affect productivity and
overall performance. Continuous use of synthetic tonic agents may cause
adverse effects; therefore, safer alternatives derived from natural products
are needed. Jambu bol leaves (Syzygium malaccense) are known to contain
various secondary metabolites that may possess tonic activity. This study
aimed to determine the tonic activity of ethanol extract of jambu bol leaves
on male white mice (Mus musculus).
Methods: This study employed an experimental design using male white mice
divided into five groups: a negative control group (0.5% Na-CMC), a positive
control group (caffeine), and three treatment groups receiving ethanol extract
of jambu bol leaves at doses of 50 mg/kgBW, 100 mg/kgBW, and 200
mg/kgBW. Tonic activity was evaluated using the natatory exhaustion method
by measuring the swimming endurance time of mice before and after 14 days
of treatment. Data were analyzed using Paired Sample t-Test, One-Way
ANOVA, and Tukey’s post hoc test.
Results: Phytochemical screening revealed that the ethanol extract of jambu
bol leaves contained alkaloids, flavonoids, tannins, saponins, and steroids,
while terpenoids were not detected. Statistical analysis showed significant
differences among treatment groups (p < 0.05). The ethanol extract was able
to increase the swimming endurance time of mice, with the best tonic activity
observed at a dose of 200 mg/kgBW, resulting in a 246.09% increase in
swimming endurance time.
Conclusion: The ethanol extract of jambu bol leaves (Syzygium malaccense)
exhibited tonic activity in male white mice (Mus musculus). The dose of 200
mg/kgBW was the most effective in enhancing physical endurance based on
swimming endurance time.
