Abstract
ABSTRAK
Latar Belakang : Daun brokoli (Brassica oleracea L.) diketahui mengandung berbagai senyawa metabolit sekunder, salah satunya adalah kumarin. Kumarin merupakan senyawa golongan benzopiron yang mempunyai beragam kegiatan farmakologis, antara lain sebagai antioksidan, antimikroba, antiinflamasi, serta antikanker. Keberhasilan ekstraksi kumarin dari bahan alam dipengaruhi oleh kesesuaian tingkat kepolaran antara senyawa dan pelarut yang digunakan. Perbedaan tingkat kepolaran pelarut dapat menyebabkan perbedaan jumlah kumarin yang terekstraksi sehingga memengaruhi kadar kumarin yang diperoleh. Oleh karena itu, dilakukan penelitian guna mendapati dampak pembeda pelarut atas kadar total kumarin pada ekstrak daun brokoli (Brassica oleracea L.) yang diekstraksi memakai metanol, etil asetat, serta n-heksana.
Metode : Penelitian tersebut ialah eksperimental yang bermaksud membandingkan kadar jumlah kumarin pada ekstrak daun brokoli (Brassica oleracea L.) yang diekstraksi menggunakan pelarut metanol, etil asetat, serta n-heksana. Penentuan kadar jumlah kumarin dilakukan memakai tekhnik Spektrofotometri UV-Vis.
Hasil : penentuan ukuran total kumarin dalam ekstrak daun brokoli (Brassica oleracea L.) menunjukkan bahwa ekstrak metanol memiliki kadar tertinggi sebesar 7,66 mg CE/g ekstrak, diikuti ekstrak etil asetat sebesar 3,92 mg CE/g ekstrak, sedangkan kadar terendah diperoleh pada ekstrak n-heksana sebesar 2,58 mg CE/g ekstrak.
Kesimpulan :
Hasil penelitian menunjukkan bahwa daun brokoli (Brassica oleracea L.) mengandung senyawa kumarin. Variasi pelarut ekstraksi memengaruhi kadar total kumarin yang diperoleh, dengan ekstrak metanol menghasilkan kadar kumarin tertinggi diperbandingkan ekstrak etil asetat serta n-heksana.

ABSTRACT
Background: Broccoli leaves (Brassica oleracea L.) are known to contain various secondary metabolites, one of which is coumarin. Coumarin is a benzopyrone compound that exhibits various pharmacological activities, including antioxidant, antimicrobial, anti-inflammatory, and anticancer properties. The efficiency of coumarin extraction from natural materials is influenced by the compatibility between the polarity of the compound and that of the extraction solvent. Differences in solvent polarity may result in variations in the amount of coumarin extracted, thereby affecting the total coumarin content obtained. Therefore, this study was conducted to investigate the effect of different solvents on the total coumarin content of broccoli leaf (Brassica oleracea L.) extracts obtained using methanol, ethyl acetate, and n-hexane.
Methods: This experimental study aimed to compare the total coumarin content of broccoli leaf (Brassica oleracea L.) extracts obtained using methanol, ethyl acetate, and n-hexane as extraction solvents. The total coumarin content was determined using the UV-Visible Spectrophotometric method.
Results: The determination of total coumarin content in broccoli leaf (Brassica oleracea L.) extracts showed that the methanolic extract had the highest coumarin content at 7.66 mg CE/g extract, followed by the ethyl acetate extract at 3.92 mg CE/g extract, while the lowest content was found in the n-hexane extract at 2.58 mg CE/g extract.
Conclusion: The results demonstrated that broccoli leaves (Brassica oleracea L.) contain coumarin compounds. Variations in extraction solvents affected the total coumarin content obtained, with the methanolic extract yielding the highest coumarin content compared to the ethyl acetate and n-hexane extracts.
