Abstract

Latar Belakang : Proses fiksasi sangat penting dalam pembuatan sediaan histologis
karena mempengaruhi kualitas struktur jaringan yang diamati. Neutral Buffered
Formalin (NBF) 10% merupakan fiksatif standar yang efektif, namun bersifat toksik
dan karsinogenik. Oleh karena itu, dibutuhkan bahan alternatif yang lebih aman dan
ramah lingkungan seperti madu, yang memiliki sifat antibakteri, antioksidan, serta
kandungan gula tinggi yang dapat menghambat degradasi jaringan. Tujuan
Penelitian: Mengetahui efektivitas madu sebagai bahan fiksatif alternatif pada
jaringan histologis hati ayam dengan konsentrasi 10%, 15%, dan 20%, serta
membandingkannya dengan NBF 10%. Metode Penelitian : Penelitian ini
merupakan eksperimen laboratorium dengan desain posttest-only control group.
Sampel yang digunakan adalah hati ayam broiler yang difiksasi menggunakan
madu dengan tiga konsentrasi berbeda dan NBF 10% sebagai kontrol. Pengamatan
kualitas jaringan dilakukan secara mikroskopis berdasarkan tampilan inti sel,
sitoplasma, dan keseragaman warna. Data dianalisis menggunakan uji ANOVA.
Hasil Penelitian : Rata-rata skor kualitas jaringan tertinggi diperoleh dari NBF
yaitu sebesar 8,8, sedangkan madu 10%, 15%, dan 20% masing-masing
menghasilkan skor rata-rata 4,76; 4,86; dan 5,16. Uji ANOVA menunjukkan
perbedaan signifikan antar kelompok perlakuan (p < 0,05). Meskipun madu 20%
menunjukkan kualitas jaringan lebih baik dibandingkan konsentrasi madu lainnya,
namun belum mencapai kualitas yang setara dengan NBF. Kesimpulan : Madu
pada konsentrasi 10–20% belum seefektif NBF 10% dalam mempertahankan
kualitas struktur jaringan histologis hati ayam. Penelitian lebih lanjut dengan
konsentrasi madu yang lebih tinggi atau waktu fiksasi yang diperpanjang
diperlukan untuk meningkatkan efektivitasnya.

Background : The fixation process is crucial in the manufacture of histological
preparations because it affects the quality of the observed tissue structure. Formalin
10% (Neutral Buffered Formalin/NBF) is an effective standard fixative, but it is
toxic and carcinogenic. Therefore, safer and more environmentally friendly
alternative materials such as honey are needed, which have antibacterial,
antioxidant, and high sugar properties that can inhibit tissue degradation.
Research Objective : To determine the effectiveness of honey as an alternative
fixative ingredient in the histological tissue of chicken liver with concentrations of
10%, 15%, and 20%, and to compare it with 10% NBF. Research Method : This
study is a laboratory experiment with a posttest-only control group design. The
sample used was broiler chicken liver fixed using honey with three different
concentrations and 10% NBF as a control. Tissue quality observation was carried
out microscopically based on the appearance of the cell nucleus, cytoplasm, and
color uniformity. Data were analyzed using ANOVA tests. Research Results : The
average highest tissue quality score was obtained from NBF, namely 8.8, while
10%, 15%, and 20% honey produced an average score of 4.76 respectively; 4,86;
and 5.16. The ANOVA test showed significant differences between treatment groups
(p < 0.05). Although 20% honey showed better tissue quality than other honey
concentrations, it did not reach the same quality as NBF. Conclusion : Honey at a
concentration of 10–20% is not as effective as 10% NBF in maintaining the quality
of histological tissue structure of chicken liver. Further research with higher
concentrations of honey or extended fixation times is needed to improve its
effectiveness.
