Abstract

ABSTRAK
Latar Belakang : Radikal bebas merupakan molekul yang dapat menyebabkan kerusakan sel dan memicu berbagai penyakit degeneratif. Salah satu upaya untuk menghambat efek radikal bebas adalah dengan menggunakan antioksidan alami. Daun sirsak (Annona muricata L.) dan umbi bit merah (Beta vulgaris L.) diketahui mengandung senyawa flavonoid, fenolik, tanin, dan betalain yang berpotensi sebagai antioksidan. Kombinasi kedua ekstrak diperkirakan mampu menghasilkan aktivitas antioksidan yang lebih baik dibandingkan ekstrak tunggal karena adanya interaksi sinergis dari senyawa aktif yang terkandung di dalamnya. Penelitian ini bertujuan untuk membandingkan aktivitas antioksidan ekstrak etanol daun sirsak, ekstrak etanol umbi bit merah, serta kombinasi keduanya menggunakan metode DPPH.

Metode : Penelitian ini merupakan penelitian eksperimental menggunakan simplisia daun sirsak dan umbi bit merah yang diekstraksi dengan metode maserasi menggunakan pelarut etanol 70%. Ekstrak yang diperoleh kemudian diuapkan menggunakan rotary evaporator hingga diperoleh ekstrak kental. Sebelum dilakukan pengujian aktivitas antioksidan, ekstrak diidentifikasi kandungan flavonoid dan fenoliknya. Selanjutnya ekstrak tunggal, kombinasi ekstrak dengan perbandingan 1:1, 1:2, dan 2:1, serta vitamin C sebagai baku pembanding dibuat dalam lima seri konsentrasi yaitu 2 ppm, 4 ppm, 6 ppm, 8 ppm, dan 10 ppm. Aktivitas antioksidan ditentukan menggunakan metode DPPH secara spektrofotometri UV-Vis berdasarkan nilai persen peredaman radikal bebas dan dihitung nilai IC₅₀ masing-masing sampel.

Hasil : Hasil penelitian didapatlan hasil berupa IC50 dari Ekstrak Daun Sirsak sebesar 19,85 ppm, Ekstrak Umbi Bit Merah sebesar 25,63 ppm, Ekstrak Kombinasi (1:1) sebesar 21,49 ppm, Ekstrak Kombinasi (1:2) sebesar 17,31 ppm, Ekstrak Kombinasi (2:1) sebesar 13,27 ppm, dan Baku Pembanding Vitamin C sebesar 5,80 ppm

Kesimpulan : Penelitian ini menunjukkan bahwa aktivitas antioksidan kombinasi Ekstrak Daun Sirsak dan Ekstrak Umbi Bit Merah (2:1) lebih baik dibandingkan ekstrak tunggalnya.

ABSTRACT
Background: Free radicals are molecules that can cause cell damage and trigger various degenerative diseases. One way to inhibit the effects of free radicals is by using natural antioxidants. Soursop leaves (Annona muricata L.) and red beetroot (Beta vulgaris L.) are known to contain flavonoids, phenolics, tannins, and betalains, which have antioxidant potential. The combination of the two extracts is thought to produce superior antioxidant activity compared to the single extracts due to the synergistic interaction of the active compounds they contain. This study aimed to compare the antioxidant activity of ethanol extracts of soursop leaves, ethanol extracts of red beetroot, and a combination of the two using the DPPH method.
Methods: This is an experimental study using crude drugs from soursop leaves and red beetroot, extracted by maceration using 70% ethanol. The resulting extracts were then evaporated using a rotary evaporator to obtain a thick extract. Prior to testing the antioxidant activity, the extracts were identified for their flavonoid and phenolic content. Furthermore, single extracts, combination extracts with ratios of 1:1, 1:2, and 2:1, along with vitamin C as a reference standard, were prepared in five concentration series: 2 ppm, 4 ppm, 6 ppm, 8 ppm, and 10 ppm. Antioxidant activity was determined using the DPPH method using UV-Vis spectrophotometry based on the percentage of free radical scavenging, and the IC₅₀ value for each sample was calculated.
Results: The IC50 values for Soursop Leaf Extract were 19.85 ppm, Red Beetroot Extract 25.63 ppm, Combination Extract (1:1) 21.49 ppm, Combination Extract (1:2) 17.31 ppm, Combination Extract (2:1) 13.27 ppm, and Vitamin C Reference Standard 5.80 ppm.
Conclusion: This study shows that the antioxidant activity of the combination of Soursop Leaf Extract and Red Beetroot Extract (2:1) is superior to that of either extract alone.
