Abstract

Latar belakang : Karies gigi merupakan salah satu masalah kesehatan gigi dan mulut yang banyak disebabkan oleh bakteri Streptococcus mutans. Daun sirih (Piper betle L.) dan daun pegagan (Centella asiatica L.) mengandung senyawa metabolit sekunder yang memiliki aktivitas antibakteri sehingga berpotensi dikembangkan sebagai bahan aktif dalam sediaan pasta gigi herbal. Penelitian ini bertujuan untuk memformulasikan dan mengevaluasi kestabilan fisik sediaan pasta gigi kombinasi ekstrak daun sirih dan daun pegagan dengan variasi konsentrasi Na-CMC sebagai bahan pengikat.
Metode : Penelitian ini merupakan penelitian eksperimental laboratorium. Ekstrak daun sirih dan daun pegagan diperoleh melalui metode maserasi menggunakan etanol 96%, kemudian diformulasikan menjadi pasta gigi dengan tiga variasi konsentrasi Na-CMC. Evaluasi kestabilan fisik dilakukan selama 28 hari pada suhu kamar dan melalui uji dipercepat (cycling test) selama 12 hari. Parameter yang diamati meliputi organoleptik (warna, bau, dan rasa), homogenitas, pH, viskositas, daya sebar, dan tinggi busa.
Hasil : Skrining fitokimia menunjukkan ekstrak daun sirih mengandung flavonoid dan tanin, sedangkan ekstrak daun pegagan mengandung alkaloid, flavonoid, tanin, dan saponin. Seluruh formula memenuhi persyaratan mutu fisik dengan nilai pH 5,35 ± 0,03–9,51 ± 0,01, viskositas 21.091 ± 272–37.749 ± 80 cP, daya sebar 2,97 ± 0,06–4,13 ± 0,31 cm, dan tinggi busa 11,0 ± 0,0–14,0 ± 1,0 mm. Setelah cycling test, seluruh formula tetap stabil, dengan Formula III menunjukkan kestabilan fisik terbaik
Kesimpulan : kombinasi ekstrak daun sirih (Piper betle L.) dan daun pegagan (Centella asiatica L.) dapat diformulasikan menjadi sediaan pasta gigi yang memiliki kestabilan fisik baik dan memenuhi persyaratan mutu selama penyimpanan.

Background : Dental caries is one of the most common oral health problems caused by Streptococcus mutans. Betel leaf (Piper betle L.) and gotu kola leaf (Centella asiatica L.) contain secondary metabolites with antibacterial activity, making them potential active ingredients for herbal toothpaste formulations. This study aimed to formulate and evaluate the physical stability of a toothpaste containing a combination of betel leaf and gotu kola leaf extracts with different concentrations of sodium carboxymethyl cellulose (Na-CMC) as a binder.
Methods: This study was a laboratory experimental study. Betel leaf and gotu kola leaf extracts were obtained by maceration using 96% ethanol and subsequently formulated into toothpaste with three different concentrations of Na-CMC. Physical stability was evaluated during 28 days of storage at room temperature and by an accelerated stability test (cycling test) for 12 days. The evaluated parameters included organoleptic properties (color, odor, and taste), homogeneity, pH, viscosity, spreadability, and foam height.
Results: hytochemical screening showed that Piper betle L. leaf extract contained flavonoids and tannins, while Centella asiatica L. leaf extract contained alkaloids, flavonoids, tannins, and saponins. All formulations met the physical quality requirements, with pH values ranging from 5.35 ± 0.03 to 9.51 ± 0.01, viscosity ranging from 21,091 ± 272 to 37,749 ± 80 cP, spreadability ranging from 2.97 ± 0.06 to 4.13 ± 0.31 cm, and foam height ranging from 11.0 ± 0.0 to 14.0 ± 1.0 mm. Following the accelerated stability (cycling) test, all formulations remained stable, with Formula III exhibiting the best physical stability.
Conclusion: A combination of betel leaf (Piper betle L.) and gotu kola leaf (Centella asiatica L.) extracts can be successfully formulated into a toothpaste with good physical stability and that meets the required quality standards during storage.
