Abstract
Latar Belakang: Media Nutrient Agar (NA) komersial yang umum digunakan di laboratorium mikrobiologi memiliki harga yang relatif mahal dan ketersediaannya sering kali terbatas. Biji nangka (Artocarpus heterophyllus) merupakan limbah pangan kaya karbohidrat dan protein yang berpotensi dimanfaatkan sebagai sumber nutrisi alternatif bagi pertumbuhan bakteri. Tujuan Penelitian: Untuk mengetahui efektivitas air rebusan biji nangka (Artocarpus heterophyllus) sebagai media pertumbuhan alternatif terhadap karakteristik morfologi dan pertumbuhan bakteri Escherichia coli. Metode Penelitian: Penelitian ini menggunakan desain kuasi-eksperimental (post-test only control group design). Pertumbuhan Escherichia coli diuji pada variasi konsentrasi media air rebusan biji nangka sebesar 10%, 20%, dan 30%, dengan menggunakan media Nutrient Agar (NA) sebagai kontrol. Parameter yang diamati meliputi jumlah koloni menggunakan metode Total Plate Count (TPC) dengan teknik spread plate, ukuran diameter koloni, serta morfologi makroskopis koloni bakteri. Analisis data dilakukan dengan uji univariat dan uji bivariat (Uji Anova) menggunakan perangkat lunak SPSS. Hasil Penelitian: Karakteristik morfologi koloni Escherichia coli pada seluruh variasi konsentrasi media alternatif (10%, 20%, dan 30%) menunjukkan karakteristik yang normal, yaitu berbentuk bulat yang seragam, permukaan halus, elevasi cembung, dengan warna putih kekuningan. Hasil analisis bivariat dengan uji Anova menunjukkan: 1) Pada ukuran diameter koloni, tidak terdapat perbedaan yang signifikan secara statistik antara kelompok media alternatif biji nangka dan media kontrol NA (p = 0,413 > 0,05). 2) Pada jumlah koloni bakteri, terdapat perbedaan yang signifikan secara statistik di antara kelompok konsentrasi media alternatif yang diuji (p = 0,001 < 0,05). Kesimpulan: Air rebusan biji nangka efektif digunakan sebagai media pertumbuhan alternatif untuk bakteri Escherichia coli. Pertumbuhan ukuran diameter koloni tidak berbeda nyata dengan media kontrol NA, namun variasi tingkat konsentrasi media alternatif secara signifikan memengaruhi jumlah koloni bakteri yang tumbuh.
Background: Commercial Nutrient Agar (NA) media commonly used in microbiology laboratories is relatively expensive and its availability is often limited. Jackfruit seed (Artocarpus heterophyllus) is a food waste rich in carbohydrates and protein which has the potential to be used as an alternative nutrient source for bacterial growth. Objective: To determine the effectiveness of boiled jackfruit seed water (Artocarpus heterophyllus) as an alternative growth medium on the morphological characteristics and growth of Escherichia coli bacteria. Methods: This study used a quasi-experimental design (post-test only control group design). The growth of Escherichia coli was tested on variations in the concentration of boiled jackfruit seed water media at 10%, 20%, and 30%, using Nutrient Agar (NA) media as a control. Parameters observed included the number of colonies using the Total Plate Count (TPC) method with the spread plate technique, colony diameter size, and macroscopic morphology of bacterial colonies. Data analysis was performed using univariate and bivariate analysis (Anova Test) via SPSS software. Results: The morphological characteristics of Escherichia coli colonies at all concentration variations of alternative media (10%, 20%, and 30%) showed normal characteristics, including a uniform circular shape, smooth surface, convex elevation, with a yellowish-white color. The bivariate analysis using the Anova test showed: 1) For colony diameter size, there was no statistically significant difference between the alternative jackfruit seed media group and the NA control media (p = 0.413 > 0.05). 2) For bacterial colony count, there was a statistically significant difference among the tested concentration groups of alternative media (p = 0.001 < 0.05). Conclusion: Boiled jackfruit seed water is effective for use as an alternative growth medium for Escherichia coli bacteria. The growth of colony diameter size did not significantly differ from the NA control media, but variations in the concentration levels of the alternative media significantly affected the number of bacterial colonies grown.
