Abstract
Latar Belakang: Pemanfaatan media kultur alternatif menjadi salah satu upaya untuk mengurangi ketergantungan terhadap media komersial yang relatif mahal. Kacang merah (Kacang Merah) memiliki kandungan protein, karbohidrat, vitamin, dan mineral yang berpotensi digunakan sebagai sumber nutrisi bagi pertumbuhan bakteri. Penelitian ini bertujuan untuk mengetahui konsentrasi minimum, rata-rata, maksimum, dan efektivitas ekstrak kacang merah sebagai media alternatif pengganti Endo Agar Plate (EAP) untuk pertumbuhan bakteri Escherichia coli.. Tujuan: Penelitian ini bertujuan untuk mengetahui pemanfaatan media alternatif kacang merah dengan variasi konsentrasi 20 %, 30 %, 40 %, 50 %, 60 %, 70 %. dan 80 %, dalam mendukung pertumbuhan bakteri Escherichia coli. Metode: Penelitian ini merupakan penelitian eksperimen yang dilakukan di Laboratorium Bakteriologi Jurusan Teknologi Laboratorium Medis Poltekkes Kemenkes Palembang. Media ekstrak kacang merah dibuat dalam tujuh variasi konsentrasi, yaitu 20%, 30%, 40%, 50%, 60%, 70%, dan 80%, dengan Endo Agar Plate sebagai kontrol. Setiap perlakuan dilakukan sebanyak empat kali pengulangan. Parameter yang diamati adalah diameter koloni Escherichia coli. Analisis data dilakukan secara univariat dan bivariat menggunakan uji One Way ANOVA setelah memenuhi uji normalitas dan homogenitas. Hasil: Hasil penelitian menunjukkan bahwa rata-rata diameter koloni pada konsentrasi 20%, 30%, 40%, 50%, 60%, 70%, dan 80% berturut-turut adalah 0,750 mm; 1,150 mm; 1,200 mm; 1,250 mm; 1,275 mm; 1,375 mm; dan 1,550 mm. Konsentrasi minimum yang mendukung pertumbuhan bakteri adalah 20%, sedangkan konsentrasi maksimum adalah 80%. Konsentrasi 70% merupakan konsentrasi yang paling efektif karena menghasilkan diameter koloni yang paling mendekati media kontrol EAP (1,372 mm). Hasil uji One Way ANOVA menunjukkan nilai signifikansi sebesar 0,024 (p<0,05), yang menunjukkan adanya pengaruh variasi konsentrasi ekstrak kacang merah terhadap pertumbuhan Escherichia coli. Kesimpulan: Media alternatif mampu mendukung pertumbuhan bakteri Escherichia coli. Di antara kelompok perlakuan alternatif, konsentrasi 70% merupakan formulasi yang paling efektif karena menghasilkan pertumbuhan diameter koloni tertinggi, meskipun belum mampu menandingi efisiensi diameter koloni pada media standar komersial EAP.
Background: The use of alternative culture media is an effort to reduce dependence on relatively expensive commercial media. Red beans (Red Beans) contain protein, carbohydrates, vitamins and minerals which have the potential to be used as a source of nutrition for bacterial growth. This research aims to determine the minimum, average, maximum concentration and effectiveness of red bean extract as an alternative medium to replace Endo Agar Plate (EAP) for the growth of Escherichia coli bacteria. Objective: This research aims to determine the use of alternative red bean media with varying concentrations of 20%, 30%, 40%, 50%, 60%, 70%. and 80%, in supporting the growth of Escherichia coli bacteria. Method: This research is experimental research conducted at the Bacteriology Laboratory, Medical Laboratory Technology Department, Health Polytechnic, Ministry of Health, Palembang. Red bean extract media was made in seven concentration variations, namely 20%, 30%, 40%, 50%, 60%, 70%, and 80%, with Endo Agar Plate as a control. Each treatment was carried out four times in repetition. The parameter observed was the diameter of the Escherichia coli colony. Data analysis was carried out univariate and bivariate using the One Way ANOVA test after fulfilling the normality and homogeneity tests. Results: The results showed that the average colony diameter at concentrations of 20%, 30%, 40%, 50%, 60%, 70%, and 80% respectively was 0.750 mm; 1,150mm; 1,200mm; 1,250mm; 1,275mm; 1,375mm; and 1,550 mm. The minimum concentration that supports bacterial growth is 20%, while the maximum concentration is 80%. The 70% concentration was the most effective concentration because it produced a colony diameter that was closest to the EAP control medium (1.372 mm). The results of the One Way ANOVA test showed a significance value of 0.024 (p<0.05), which indicated the influence of variations in the concentration of red bean extract on the growth of Escherichia coli. Conclusion: Red bean alternative media is able to support the growth of Escherichia coli bacteria. Among the alternative treatment groups, the 70% concentration was the most effective formulation because it produced the highest colony diameter growth, although it was not able to match the efficiency of colony diameter on standard commercial EAP media.
