Abstract

Latar Belakang: Sitologi mukosa mulut merupakan metode pemeriksaan untuk mengamati morfologi sel epitel rongga mulut. Salah satu faktor yang dapat memengaruhi kualitas sediaan sitologi adalah proses pengeringan. Perbedaan suhu pengeringan diduga memengaruhi kualitas morfologi inti sel, sitoplasma, dan latar belakang pada sediaan yang diwarnai menggunakan Giemsa. Tujuan Penelitian: Untuk mengetahui pengaruh variasi suhu pengeringan sediaan sitologi mukosa mulut terhadap hasil pewarnaan Giemsa. Metode Penelitian: Penelitian eksperimental ini menggunakan 16 sampel yang dibagi menjadi dua kelompok, yaitu pengeringan pada suhu kamar dan hot drying oven suhu 45°C. Kualitas sediaan dinilai berdasarkan inti sel, sitoplasma, dan latar belakang. Data dianalisis secara univariat dan bivariat menggunakan uji Wilcoxon. Hasil Penelitian: Pengeringan pada suhu kamar menghasilkan rerata skor kualitas sediaan sebesar 4,94 dengan persentase kualitas sediaan baik sebesar 100%, sedangkan pengeringan menggunakan hot drying oven suhu 45°C menghasilkan rerata skor sebesar 4,50 dengan persentase kualitas sediaan baik sebesar 93,8%. Hasil uji Wilcoxon menunjukkan nilai p = 0,071, sehingga tidak terdapat perbedaan yang signifikan antara kedua metode pengeringan. Kesimpulan: Variasi suhu pengeringan tidak berpengaruh terhadap kualitas sediaan pewarnaan Giemsa sitologi mukosa mulut. Pengeringan pada suhu kamar maupun hot drying oven suhu 45°C sama-sama menghasilkan sediaan dengan kualitas baik, dengan persentase kualitas sediaan baik masing-masing sebesar 100% dan 93,8%.

Background: Oral mucosal cytology is an examination method used to observe the morphology of oral epithelial cells. One of the factors that may affect the quality of cytological preparations is the drying process. Variations in drying temperature are presumed to influence the quality of nuclear morphology, cytoplasm, and background in preparations stained with Giemsa. Research Objective: To determine the effect of drying temperature variations on the quality of oral mucosal cytology preparations stained with Giemsa. Research Methods: This experimental study used 16 samples divided into two groups, namely drying at room temperature and drying using a hot drying oven at 45°C. Preparation quality was assessed based on nuclear, cytoplasmic, and background characteristics. Data were analyzed using univariate and bivariate analyses with the Wilcoxon test. Research Results: Drying at room temperature produced a mean preparation quality score of 4.94, with 100% of the preparations categorized as good quality. In contrast, drying using a hot drying oven at 45°C produced a mean score of 4.50, with 93.8% of the preparations categorized as good quality. The Wilcoxon test showed a p value of 0.071, indicating no statistically significant difference between the two drying methods. Conclusion: Variations in drying temperature did not affect the quality of oral mucosal cytology preparations stained with Giemsa. Both drying at room temperature and drying using a hot drying oven at 45°C produced preparations with good quality, with percentages of good quality preparations of 100% and 93.8%, respectively.
